Healthy and diseased human lung model

The aim of this study was to develop human models of healthy lung and chronic obstructive pulmonary disease (COPD) to accelerate the development of new therapeutics for lung diseases.
Five components were used and optimized: (i) cell type, (ii) membrane structure, (iii) environmental conditions, (iv) cellular arrangement, and (v) matrix structure and composition. Models were initially established using immortalized human cell lines, before cell lines were replaced by primary cells, thereby enabling the production of patient-specific models. Cellular barrier function, permeability, tight junctions, tissue structure, tissue composition, and cytokine secretion were investigated.
The model can be utilized to test safety, and efficacy of new therapeutics as well as to test toxicity and injury induced by inhaled pollution or pathogens.
