Immunocompetent human kidney on-a-chip model to study renal inflammation

Kidney damage and dysfunction is a growing global health concern, causing high morbidity and mortality rates. Many renal diseases are driven by the immune system, but developing targeted therapies has been challenging due to insufficient understanding of underlying mechanisms. This study establishes a human immunocompetent 3D in vitro co-culture model of the proximal tubule in a high-throughput microfluidic platform to study renal functionality and inflammatory processes. The model successfully incorporates renal proximal tubule epithelial cells, human umbilical vein endothelial cells, and human primary monocytes, allowing for the study of renal inflammation. The platform demonstrates potential for studying renal functionality in health and disease, as well as for drug screening, due to its compatibility with automation and high throughput capabilities.
