Cholangiocyte organoids to study drug-induced injury

Drug-induced bile duct injury is a frequently observed clinical problem with a wide range of pathological features. In this study, the mechanisms of chlorpromazine (CPZ) induced bile duct injury were investigated using advanced in vitro cholangiocyte cultures, generated from intrahepatic cholangiocyte organoids (ICOs). CPZ disrupted the stability of the cell layer by reducing key cell-connecting proteins, leading to a loss of barrier function and increased cell layer permeability. It also caused oxidative stress and reduced the levels of bile acid transport proteins, leading to bile acid accumulation. These results show that ICOs present a model to identify toxic drugs affecting the bile ducts while providing mechanistic insights into hepatotoxicity.
