Optimization of zebrafish spheroid production

Fish cell spheroids are promising 3D culture models in ecotoxicology. The aim of this study was to optimize conditions for fish cell spheroid formation with regard to spheroid quality, and fast spheroid formation.
Therefore, the effectiveness of the traditional hanging drop (HD) and orbital shaking (OS) spheroid formation was accessed and compared. Modifications in cell density, culture duration, pretreatment of culture dishes, format of well-plates, and combined use of both methods (HD+OS) were also investigated. Two different zebrafish cell lines were used, a zebrafish epithelial liver cell line (ZFL), and a zebrafish embryo cell line (ZEM2S).
It was found that the OS method in 96-well ultralow attachment-plates is particularly useful because it forms a great number of single spheroids in only one step. This study contributes to fast, reproducible, and simple piscine spheroid formation and supports the application of fish 3D models in industry and academia.
