Optimization of a colon model

The commercially available RepliGut® Planar model was developed to replicate human colonic epithelium in a monolayer format. The aim of this study was to optimize the system with regard to variability across cells from multiple human donors, cell lots, and passage numbers. Colon intestinal epithelial cells were derived from intestinal tissue obtained from 4 donors post-mortem. Assessment criteria included barrier formation and integrity, gene expression, and cytokine responses. It was shown that control of cell passage number reduces variability and maximizes physiological relevance of the model. After optimizing passage number, donor specific differences in cytokine responses were demonstrated, suggesting biologic variability is observable in cell cultures derived from multiple human sources. The findings highlight key considerations for designing primary-cell derived assays, as well as establish the utility of the RepliGut® Planar platform for robust development of predictive drug-response assays.
