Human gut epithelium-on-a-chip

In this study an innovative organ-on-a-chip device, named MINERVA 2.0, is demonstrated. The device is optically accessible and allows combination of several devices by a serial connection.
A gut epithelium-on-a-chip model was developed by cultivation of human epithelial cells (Caco-2) under perfusion. The cells formed a three-dimensional layer with features of the gut epithelium including tight junctions. The formed epithelium was investigated using trans-epithelial resistance and FITC-dextran permeability, mucin-2 expression, and transcriptomic profile was assessed by RNASeq.
The results suggest that the developed gut-on-a-chip model displays key gut epithelium features and could be the basis to build customizable multiorgan-on-a-chip platforms.
