Testing the efficacy of topical psoriasis treatments in a skin inflammation model

In this study, the effects of two topical treatments for psoriasis were investigated. T-helper 17 (Th17) cytokines play a crucial role in psoriasis pathophysiology by triggering inflammatory responses that lead to skin changes. Proteins marking epidermal differentiation are dysregulated in psoriasis. A fixed combination of Calcipotriol/Betamethasone dipropionate (Cal/BD) foam and Clobetasol propionate (CP) cream is a common treatment. The study utilized a human Th17 skin inflammation model (InflammaSkin®) to assess the impacts of these treatments on Th17 cytokines and epidermal differentiation. Cal/BD foam was applied once or twice daily. It strongly inhibited Th17 cytokine production and regulated the expression of Loricrin (LOR) and Involucrin (IVL), but not Filaggrin (FLG), at mRNA and protein levels. CP cream reduced Th17 cytokines, though less effectively than Cal/BD foam, and had no impact on the expression of differentiation markers. When compared to CP cream, the Cal/BD foam combination demonstrated better suppression of Th17 cytokines and improved epidermal differentiation. This resulted in an overall stronger enhancement of the skin. These findings contribute to understanding the efficacy of Cal/BD foam, supporting its use for long-term treatment of psoriasis vulgaris.
