In vitro assay for assessment of vaccine-induced inflammasome activation

The aim of this study was to develop and evaluate a quantitative in-vitro assay for inflammasome (multi-protein complexes of the innate immune system) activation by aluminium-containing vaccines. Therefore, the effects of Diphtheria-Tetanus-Pertussis combination vaccines from two manufacturers and their respective aluminium adjuvants, aluminium phosphate and aluminium hydroxide were evaluated in an in-vitro assay based on human monocytes and their interleukin production. All vaccines and adjuvants tested showed a dose-dependent increase in interleukin-1β production and a concomitant decrease in cell viability, suggesting inflammasome activation. Inflammasome activation may be used to measure adjuvant biological activity as an important quality attribute for the control or characterization of the adjuvant.
