Human full thickness skin model for drug testing

In this study, an open-source full-thickness skin model (FTSm) with an in vivo-like architecture and optimized co-culture of fibroblasts and keratinocytes is described and its potential for drug testing is assessed. The FTSms were developed using endogenous extracellular matrix to recreate the dermal compartment, avoiding animal-derived hydrogels. The integrity of the skin barrier was analysed by challenging the surface with detergents and skin irritation studies were performed based on OECD guidelines. The FTSm demonstrated structural and barrier properties comparable to the native human skin and presented better barrier properties than commercial models. In the future, these models could help speed up the translation of a new candidate therapeutics to the clinic and/or the market.
