SARS-CoV-2 in human intestinal organoids

This study investigated the infection of human-pluripotent stem cell-derived intestinal organoids by SARS-CoV-2. The authors also tested three drugs (remdesivir, famotidine and EK1) for their capability to prevent infection of the cells. Infection of host cells by SARS-CoV-2 is known to require angiotensin-converting enzyme 2 (ACE2) and transmembrane serine protease 2 (TMPRSS2). These were expressed in almost all differentiated cell types of the organoid model except for goblet cells. This finding is mirrored in the susceptibility to infection as SARS-CoV-2 could infect and replicate in all cell types (except goblet cells) leading to organoid deterioration. Of the three drugs tested, remdesivir and EK1 blocked the infection of intestinal organoids, but not famotidine.
