h-CLAT activation test for skin sensitization
Regulatory accepted 2008
European Union Reference Laboratory for Alternatives to Animal Testing, Ispra, Italy
The h-CLAT discriminates between sensitizing and non-sensitizing chemicals. The human monocytic leukemia cell line THP-1 cells used are derived from monocytes which show similar reactions to dendritic cells after exposure to skin sensitizing chemicals, such as upregulation of surface markers and cytokine production. The (change in) expression of these markers is determined and the skin sensitizing potential of a chemical is derived from this.
Validated and regulatory approved under TM2008-05 (EU) by EURL ECVAM (TSAR list), OECD Test No. 442E.
