KeratinoSens assay for the testing of skin sensitizers
Regulatory accepted 2010
European Union Reference Laboratory for Alternatives to Animal Testing, Ispra, Italy
The KeratinoSens™ test method supports discrimination between sensitizing and non-sensitizing chemicals. The activation of a signalling pathway, which occurs during a key event of the Adverse Outcome Pathway (AOP) of skin sensitization, is assessed.
The KeratinoSens™ is performed with an immortalized, adherent human HaCaT keratinocyte cell line containing a plasmid with the luciferase gene. Quantitative optical measurement is used to indicate small electrophilic substances that may act as skin sensitizers.
Validated and regulatory approved under TM2010-03 (EU) by EURL ECVAM (TSAR list), OECD Test No. 442D.
