Microbiome integration into a microfluidic gut-model

This study describes the establishment of a microfluidic-based gut model which allows the co-culture of human and microbial cells. The application of fluidic shear stress enhanced the cellular differentiation and barrier formation of the intestinal cells. The microfluidic system enables the perfusion-based cell culture with microbiomes and analyses of the effect of microbiomes on intestinal epithelial barrier functions. Additionally, a disease model of lipopolysaccharide (LPS)-induced intestinal injury and inflammation was described and the therapeutic effect of probiotics was assessed. This gut-on-a-chip could provide insights into the interaction between the intestinal microbiome and human health and disease. Moreover, it is a potentially powerful tool for pharmaceutical applications.
