3D organ-on-a-chip system with automatic perfusion

Organ-on-a-chip technologies are becoming increasingly interesting for drug development and personalised medicine. However, there are still several features of these systems that are not fully used and, current setups are based on single tissues and rely on manual handling, which limits the expansion of this technology in the industry. Here, human primary fibroblasts and adipose-tissue derived stem cells were used in a customised thermoplastic disc to develop a 3D model that takes advantage of a fully automatised setup. The results showed that it was possible to seed 20 tissue chambers where cells formed 3D constructs and, using low-speed rotation, the discs could be perfused without vasculature-like structures. Overall, the researchers propose an organ-on-a-disc model that allows highly customisable conditions for cell culture with automatic centrifugal perfusion, making it a potential tool for high-throughput assay systems for drug development.
