LuSens assay for skin sensitization analysis
Validated Method 2011
European Union Reference Laboratory for Alternatives to Animal Testing, Ispra, Italy
The LuSens test detects skin sensitizing and non-sensitizing chemicals. The concept is based on the KeratinoSens™ adopted by the OECD.
The test measures the activation of a signalling pathway that is triggered under a key event of the Adverse Outcome Pathway (AOP) of skin sensitization.
Genetically modified keratinocytes derived from HaCaT cells carrying the luciferase gene are used for the test. The upregulation of luciferase activity, which is optically measured, is an indicator of electrophilic stress caused by cysteine-reactive substances.
Validated and regulatory approved under TM2011-10 (EU) by EURL ECVAM (TSAR list), OECD Test No. 442D.
