Influence of cigarette smoke on mitochondrial metabolism in bronchial cells

Exposure to cigarette smoke (CS) is the major risk factor for the development of chronic obstructive pulmonary disease. The effects of CS exposure on molecular mechanisms involved in mitochondrial quality control in airway epithelial cells are incompletely known. In this study, the occurrence of key regulatory molecules involved in mitochondrial metabolism was investigated in different in vitro models of CS exposure using human primary bronchial epithelial cells (PBEC) from healthy individuals and patients with chronic obstructive pulmonary disease (COPD). These cells were either cultured in an undifferentiated state and exposed to WCS (cigarette smoke) or CSE (cigarette smoke extract), or they were differentiated by culture in an air-liquid culture system (ALI) and subsequently exposed. In addition to assessing the acute response of mitochondrial quality control processes to CS exposure, the effects of repeated/chronic WCS exposure during differentiation and mitochondrial recovery after cessation of WCS exposure were also examined in our ALI-PBEC model (undifferentiated PBEC cultured in ALI to induce differentiation). The results showed that smoke exposure alters the regulation of mitochondrial metabolism in airway epithelial cells, but the observed changes may differ between the different culture models used.
