Skin equivalent suitable for 96-well plates and organ-chips

The aim of this study was to develop a full-thickness skin equivalent (ftSE) with the size of a 96-well cell culture insert that can be used for an organ-on-a-chip system and high throughput assays. Human dermal fibroblasts and keratinocytes were used for the assembly of the ftSE. The immunological staining showed regular epidermal development and expression of typical marker proteins indicating an in vivo-like differentiation process. First tests on a two-organ-chip with fluorescein showed promising results for the application of the ftSE in substance penetration studies.
