Patient-specific model of aortic aneurysms

Amsterdam University Medical Centers, Amsterdam, Netherlands
In this study, smooth muscle cells derived from the aortic wall of patients with abdominal aortic aneurysms were seeded on polymeric scaffolds. Human endothelial cells were introduced after 5 weeks of culture. The spatial organization of the smooth muscle cells was monitored as well as the formation of extracellular matrix (ECM). The model exhibits viscoelastic properties comparable to biological vessels and cytoskeletal organization increases during the 5 weeks in culture. Increased cytoskeletal alignment and decreased ECM production indicate the different organization of aortic aneurism patients’ cells compared with control. The model can be used for translational studies of smooth muscle cells and their role in aortic aneurysms.
