New liver-on-a-chip model for improved drug testing

There is a lack of liver-on-a-chip models that can reliably predict both metabolism and transport of drugs in humans. In this work, a novel system based on human liver organoids and special hollow fiber membranes was developed. The organoids formed well-matured, liver cell-like layers on these membranes, which showed important enzymes for drug degradation and transport proteins. The system made it possible to study the degradation and distribution of test substances in different areas of the chip. In addition, the system was successfully coupled with an artificial intestine to simulate the absorption of drugs. This developed liver-on-a-chip model represents a promising approach to better predict the pharmacokinetics of drugs in humans.
